Multilevel Illumination Coding for Fourier Transform Interferometry in Fluorescence Spectroscopy

چکیده
Fourier Transform Interferometry (FTI) is an interferometric procedure for acquiring HyperSpectral (HS) data. Recently, it has been observed that the light source highlighting a (biologic) sample can be coded before the FTI acquisition in a procedure called Coded Illumination-FTI (CI-FTI). This turns HS data reconstruction into a Compressive Sensing (CS) problem regularized by the sparsity of the HS data. CI-FTI combines the high spectral resolution of FTI with the advantages of reduced-light-exposure imaging in biology. In this paper, we leverage multilevel sampling scheme recently developed in CS theory to adapt the coding strategy of CI-FTI to the spectral sparsity structure of HS data in Fluorescence Spectroscopy (FS). This structure is actually extracted from the spectral signatures of actual fluorescent dyes used in FS. Accordingly, the optimum illumination coding as well as the theoretical recovery guarantee are derived. We conduct numerous numerical experiments on synthetic and experimental data that show the faithfulness of the proposed theory to experimental observations.

Holographic interferometry and the fractional Fourier transformation.
The fractional Fourier transform (FRT) is shown to be of potential use in analyzing the motion of a surface by use of holographic interferometry. The extra degree of freedom made available by the use of the FRT allows information regarding both translational and tilting motion to be obtained in an efficient manner.
متن کاملGroup delay measurement of fiber Bragg grating resonances in transmission: Fourier transform interferometry versus Hilbert transform
Several methods exist to measure the group delay of a fiber Bragg grating. Here, we compare two such methods, namely the Hilbert transform (HT) of the device transmission spectrum and standard Fourier spectral interferometry. Numerical simulations demonstrate that both methods work not only for ideal, lossless devices but also for ones with realistic absorption. Experimental measurements show t...
متن کاملMultichannel Fourier Transform Raman Spectroscopy: Combining the Advantages of CCDs with Interferometry
A common-path (Sagnac) interferometer combined with a chargecoupled device (CCD) was evaluated for Raman spectroscopy in the near-infrared region. A spatial interferogram of the scattered light was projected onto the face of the CCD, and a Fourier transform of the intensity vs. pixel data yielded a Raman spectrum. This mult ichannel Fourier transform (MCFT) technique retains several advantages ...
متن کاملSuperresolved swept-wavelength interferometry using frequency estimation methods.
The high signal-to-noise ratios typical of swept-wavelength interferometry (SWI) enable distance measurements to be superresolved with 2σ uncertainties as low as 10-4-10-5 of Fourier transform-limited resolution. We compare three methods of superresolving SWI distance measurements: Local Linear Regression (LLR), Estimation of Signal Parameters via Rotational Invariance Tec...
متن کاملTomographic diffractive microscopy of transparent samples
We report a tomographic diffractive microscope, which permits imaging non-labelled transparent or semi-transparent samples. Based on a combination of microholography with a tomographic illumination, our set-up creates 3-D images of the index of refraction distribution within the sample. One acquires successively interferograms, rotating the illumination (the specimen being static) and using pha...
